Pyrogen-free buffers, acid or basic solutions

Buffer solutions, HCl and NaOH for resolving interferences and for sample preparation.

 

Pharmaceutical or biotechnological products may contain interfering factors that prevent the success of the LAL test. Some have a pH outside the optimal 6.0-8.0 range, or contain chelating agents or excess ions, proteins or polysaccharides.

In most cases, interferences can be avoided or limited by using specific buffers and pyrogenic HCl or NaOH solutions.

Endotoxin-free Tris buffers are particularly suitable for performing LAL on products that exhibit pH interference. The lysate can be dissolved in Tris buffer instead of water (according to the lysate manufacturer's specifications). Therefore the uncolored buffer is used to dissolve the lysates for the kinetic method while the colored buffer is used to reconstitute the lysates for the gel-lot. In this way it is often possible to avoid adjusting the pH of the product under test.
