qPCR Method

PCR (Polymerase Chain Reaction) is a method used to amplify a specific DNA sequence in a reaction containing, among other components, a thermostable DNA polymerase, nucleotides, and primers complementary to the target sequence.

The methodology involves several steps:

Denaturation, annealing, and extension – The DNA molecule denatures when the solution is heated, separating into two strands. Upon cooling, primers bind to the target sequences on the separated DNA strands, and DNA polymerase synthesizes a new strand by extending the primers with nucleotides, creating a copy of the DNA sequence (amplicons).
Amplification – This cycle of denaturation, annealing, and extension is repeated, exponentially increasing the number of target amplicons.
Measurement – In real-time PCR, the signal is measured at each cycle, typically using specific fluorescent probes. Fluorescence is detected by a reader, and associated software plots fluorescence intensity against cycle number, allowing determination of the presence or absence of the target organism.
